Obiettivi formativi

Il C.I. contribuisce alla formazione degli studenti nell'Area Biotecnologica del CdS. Il modulo ha lo scopo di fornire competenze su: tecniche di coltura in vitro utili per il miglioramento delle specie vegetali; i principali aspetti applicativi delle biotecnologie con particolare riferimento al miglioramento genetico e allo studio delle specie vegetali con enfasi su quelle arboree da frutto; le basi genetiche dei caratteri di interesse e delle principali strategie biotecnologiche per ottenere piante con caratteri migliorati di interesse agroindustriale.

 

 

The Integrated Course contributes to the formation activities of the Biotechnological Area of education of the Degree in Plant Biotecnologies. This class has the aim of providing knowledge and competence on: techniques of in vitro culture of plants; the main practical aspects of biotechnologies with particular attention to breeding and studying fruit and nut crops; the genetic bases of traits of interest and of the main strategies to obtain plants with improved characters of agroindustrial interest.

Attraverso le attività formative del C.I. i laureati acquisiranno competenze teoriche e operative relative agli aspetti applicativi delle biotecnologie che interessano le piante di interesse agroalimentare ed industriale. In questo modulo, in particolare, lo studente acquisirà conoscenze sulle metodologie sperimentali e sugli obiettivi della coltura in vitro e della trasformazione genetica vegetale. Lo studente acquisirà, inoltre, conoscenze sulle applicazioni delle biotecnologie al miglioramento genetico e allo studio delle specie vegetali, con particolare riferimento alle specie arboree da frutto.

 

Through the educational activities of this Integrated Course, the degree holders will have theoretical and practical skills related to applied aspects of biotechnology involving plants of agri-food and industrial interest. In this class the student will acquire knowledge on the experimental methods and objectives of the in vitro culture and genetic transformation of the plant. Furthermore, the student will acquire knowledge on the applications of biotechnologies to breeding and to the study of plants of interest in the agri-food industry with particular reference to woody nut and fruit species.

Modalità di verifica dell'apprendimento

L'apprendimento viene verificato attraverso la discussione con la classe degli argomenti trattati alla fine di ogni lezione o all'inizio di quella successiva in modo tale da evidenziare i contenuti più rilevanti. Le capacità di applicare conoscenza e comprensione e le abilità comunicative verranno sviluppate e verificate attraverso la presentazione di lavori di gruppo.

Learning is verified by discussing with the class the topic presented during the lesson, underlining the most relevant aspects of each one. The applying knowledge and understanding skills and the communication skills will be developed and evaluated by the presentation of group works.

Programma

Introduzione alla materia: principali applicazioni, sviluppi e prospettive delle biotecnologie per le piante e in particolare per le specie arboree da frutto; obiettivi e problematiche di miglioramento genetico delle specie arboree da frutto.

Tecniche di coltura in vitro: principi generali, micropropagazione, morfogenesi in vitro, coltura di protoplasti, fusione somatica, ottenimento di aploidi.

Basi genetiche e modificazioni dell’habitus vegeto-produttivo della pianta; interventi che influenzano le dimensioni della pianta o che permettono il superamento della giovanilità e delle barriere di incompatibilità.

Basi genetiche della qualità e modificazioni per migliorare le caratteristiche del frutto per quanto riguarda: quadro aromatico, contenuto in vitamine A, C ed E, colore, tessitura, contenuto in acidi organici e in zuccheri, composizione in acidi grassi, apirenia, allergeni. Interventi per variare l’andamento della maturazione del frutto: riduzione e soppressione della sintesi di etilene.

Esercitazioni di laboratorio: analisi al sequenziatore semiautomatico con elaborazione dei dati; trasformazione genetica di una specie arborea.

 

 

 

Introduction to the subject: the main applications, developments and prospects of biotechnology in plants and fruit tree species.
Techniques of in vitro culture: general principles, micropropagation, in vitro morphogenesis, protoplast culture, somatic fusion, obtaining and using haploids.

Genetic bases and modifications of the vegetative and productive tree habit; interventions and studies for overcoming self-incompatibility and juvenility and for changing tree habit and rooting ability.

Genetic bases and modifications for improving fruit and nut quality traits: aroma, vitamin content, pulp softening, organic acid and sugars content, fatty acids composition, colour, seedlessness. Allergenes in fruits and nuts. Genetic bases and modifications for changing the ethylene methabolism and the ripening process.

Practical activities: analysis using an authomated sequencer, data elaboration; genetic transformation of a tree species
